Semax (5mg) — Research Peptide

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from the ACTH(4-10) fragment of adrenocorticotropic hormone. The Pro-Gly-Pro C-terminal extension was added to improve metabolic stability compared to the native ACTH fragment. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Semax has undergone clinical investigation in Russia for neurological applications and has been registered as a pharmaceutical for conditions including ischaemic stroke and cognitive impairment.

Mechanism of Action

Semax’s primary documented mechanisms involve modulation of the brain-cortex neurotrophin system. Published research demonstrates Semax upregulates expression of BDNF (brain-derived neurotrophic factor), NGF (nerve growth factor), and their receptors TrkB and TrkA in rat cortical and hippocampal tissue. These neurotrophins are critical for neuronal survival, synaptic plasticity, and cognitive function, making Semax of interest in neuroplasticity research.

Semax also activates dopaminergic and serotonergic systems, documented through changes in monoamine turnover in rodent models. Unlike ACTH, Semax does not significantly stimulate cortisol release at research concentrations — suggesting its CNS effects are independent of HPA axis activation, possibly mediated through melanocortin receptors in the brain.
